Functionally, both of these effects for Assassination should be 100% uptime (after the initial Vile Tincture stacks). This makes for an incredibly boring and uneventful tier set bonus. The current chance to grant you Vile Tincture is set at 8%. This honestly feels like a typo. In single target, Garrote and Rupture combined deal damage slightly more than once a second. That would lead to needing an average of roughly 2 minutes to reach max stacks, but the buff itself is 8 seconds.
