After defeating each of the seven demi-god bosses in , players are granted a Great Rune, unique items that grant powerful buffs to players once activated. The first time a player will likely encounter one will be following the fight with Godrick, The Grafted, in Stormveil Castle.
However, it is not enough to acquire these runes, as they will be useless until further steps are taken to activate them. Following the acquisition of each Great Rune, players will need to scope out a corresponding Divine Tower, where they will activate it, and then need to use a Rune Arc to receive its benefits, which will remain active until death.
To use Godrick's Great Rune, Elden Ring players must ascend a Divine Tower, and to use the Remembrance of the Grafted, players must meet with an NPC.

This rune will raise all attributes by five.
Great Rune of the Unborn
This rune can be obtained by defeating Rennala, Queen of the Full Moon, in the Raya Lucaria Academy in Liurnia of the Lakes.
This Great Rune is unique, as it cannot be equipped or activated. Instead, it will allow players to respec stats in , in the Reya Lucaria Grand Library, in exchange for a Larval Tear.
Radahn's Great Rune
This item can be obtained by defeating Starscourge Radahn in Redmane Castle in Caelid.
Radahn's Great Rune will raise a player's Health Points by 15%, Stamina by 12.5%, and Focus Points by 12.5% once activated.

Acquired by killing Morgott, the Omen King in Leyndell, the Royal Capital.
Activating this Great Rune will increase your total Health Points by 25%.
Rykard's Great Rune
You must slay Rykard, Lord of Blasphemy, in the Volcano Manor in Mt. Gelmir.
Activating this Great Rune will allow players to heal themselves by 185 points every time they have slain an enemy.
