You must defeat a powerful Regenerador in the Incubation Lab. This Blue Request Note is located just to the left of the control panel accessible directly after Ashley creates a hole in the wall using a wrecking ball. Once you've got the Blue Request Note in your inventory, open up the gate opposite so you can easily access the part of the Island you need.
