Caryll Runes are upgrades in Bloodborne that give your character enhancements, skills, and various traits, allowing them to get the edge in battle. If you played the Dark Souls titles before this, they are similar to the Rings found in those titles.

These Runes allow you to do a wide range of things, such as earning more Blood Echoes when defeating enemies, transforming into a beast, carrying more Blood Vials, and much more. In Bloodborne, every slight advantage you can get, you should take as it can open the door for insane builds that can rock even the most formidable foes.
Before you can equip Caryll Runes in Bloodborne, you must first meet a few prerequisites. First, you will need to obtain the Rune Workshop Tool, which is located in the room that becomes available after you defeat the Witch of Hemwick boss located in Hemwick Charnel Lane.
After obtaining the Rune Workshop Tool, you can then equip Caryll Runes via the Memory Altar in the Hunter's Dream. You can equip three Runes at a time but cannot equip multiple of the same Rune tier. For example, Clawmark 1 + Clawmark 1 + Clawmark 1 is not possible. However, if you have different tiers of the same Rune type, those are stackable. For example, Clawmark 1 + Clawmark 2 + Clawmark 3 is possible.
