Fans commonly say that DC Comics' version of Thanos is Darkseid, which is a fitting comparison, but it would be more accurate to say that a forgotten Superman villain is DC's version of the Mad Titan. Thanos is already a powerhouse without the Infinity Gauntlet; he has super strength, speed, durability, and can manipulate energy and matter. With the Infinity Gauntlet, he can essentially do anything. Even at his strongest, Darkseid has never reached anything near omnipotence.
Most fans don't know this, but Thanos cannot use the Infinity Gauntlet past the boundary of Earth-616. While he is one of the most omnipotent supervillains in the Marvel Universe, Thanos can only alter space, time, reality, and all the rest, in one universe. Furthermore, Marvel's Illuminati has nullified Thanos' element of surprise; they keep a constant watch on the Infinity Gems to make sure Thanos never reaches his most dangerous state again. Unlike Thanos, one overlooked and underutilized Superman villain has the power to do absolutely anything and isn't limited to one universe.

Even though Mr. Mxyzptlk usually isn't considered one of Superman's most dangerous threats, his capability is similar to those offered by Marvel's Infinity Gauntlet; in fact, his magical energy has fewer restrictions. Mxyzptlk is a mischievous and omnipotent imp from the Fifth Dimension who can do anything imaginable, like Thanos can with the Infinity Gauntlet.
