To unlock this door, you'll need to defeat the two brothers that can be found later in the level besides a locked door, just after you've cleared the room full of guys laundering drug money. This Bunch of Keys will also unlock the gate next to them, which is necessary to keep moving forward through the level, so it's unmissable.

Taking this shortcut will skip you all the way the gym, where you can choose to face a room full of fairly simple enemies or move on to the second of the trials.
To unlock this shortcut, you'll need to defeat the level's first mini-boss, who can be found by following the level's normal path.
