In Cult of the Lamb, you'll encounter Tarot Cards when you go on crusades. They'll provide you with a range of bonuses, from extra health to a bump in reward quality to giving you defensive bonuses that make your runs easier. To get them, you usually need to find card rooms during your crusade, which are only found on nodes with combat. Sometimes, you can acquire them as room rewards.

You'll begin each crusade with a clean slate - every run will provide you with a new set of Tarot Card rewards. There are 36 Tarot Cards in the game, and you'll have to unlock the vast majority of them through quests or various merchants.
Some of these cards are purchased in certain areas that must be unlocked first. Here's how to unlock them.
Once you've beaten Ratau at Knucklebones, you'll now be able to encounter the next Knucklebones opponent during a crusade. Only once you've beaten this character can you face the next one.
Sometimes, when given the choice to pick up a Tarot Card, you'll be presented with either shiny or rainbow-colored cards. These are stronger versions of the cards that have either one or two + symbols next to their names. You will usually be able to see just how much stronger they are.
These are very rare - do not rely on encountering them.
